Macdonald began his performances at stand-up clubs in Ottawa. In 1987, he appeared at the Laughs Comedy Festival in Montreal. In 1993, he joined the cast of NBC's Saturday Night Live and worked there until he was fired in 1998A year later, he was asked to host the show again.

His other shows/appearances include Weekend Update, CBS's Late Show with David Letterman, The Norm Show, Norm Macdonald Live (2013-2017), The Jackie Thomas Show (1993), The Larry Sanders Show (1995), The Drew Carey Show, among others.

He has confessed that he is a gambler and stated that his gambling addiction began after he won a six-figure win at a craps table in Atlantic City. In a 2011 interview, Macdonald revealed that he had lost all his money gambling three times. He added that the largest amount he lost was $400,000!

On March 30, 2011, he appeared on The Howard Stern Show, where he made several gay jokes for which he received several criticisms. He was asked if he was at all uncomfortable with homosexuality, and he replied that he was not only a supporter of gay rights but "fascinated" by gay sexual practice.
